MetaVia Inc. (Nasdaq: MTVA) announced on 5 November 2026 that its experimental drug DA-1726 produced a 68% placebo-adjusted reduction in liver fat after 16 weeks of once-weekly treatment, alongside 13.5% placebo-adjusted weight loss and a 0.24 percentage-point placebo-adjusted drop in HbA1c. The company said the interim Phase 1 Part 3B results were presented on 4 November 2026 at the Precision Liver Symposium in Denver. All five DA-1726-treated participants who entered with liver steatosis achieved MRI-PDFF below 5% by Day 109.
Context — why this matters now
The report frames the interim readout as the strongest clinical evidence MetaVia has generated for DA-1726. The company's chief executive, Hyung Heon Kim, said the data "provide a strong biological and clinical foundation for this next stage of development," pointing to the drug's dual GLP-1 and glucagon receptor mechanism and its concentration in liver and adipose tissue observed in preclinical studies. That combination is the basis for the company's plan to move into metabolic dysfunction and alcohol-associated liver disease (MetALD) and alcohol-associated liver disease (ALD).
The company cited its own estimate that 10.7 million U.S. adults have MetALD, a condition defined by a steatotic liver together with at least one cardiometabolic risk factor. MetaVia said there are currently no approved therapies specifically for these patient populations, which is the commercial gap the program is targeting.
The trigger for the announcement was the symposium presentation, not a regulatory filing or a financing event. MetaVia also said it will host an invitation-only key opinion leader dinner during the American Association for the Study of Liver Diseases' The Liver Meeting 2026, focused on MetALD and the evolving treatment landscape.
The company gave no prior-period comparison for the same endpoints in earlier parts of the Phase 1 trial, and it did not disclose the terms of any partnership, licensing arrangement or financing tied to the data. Those gaps matter because the readout is interim, drawn from a small sample, and generated in otherwise healthy adults with obesity rather than in patients with diagnosed liver disease.
Data — what the numbers show
The interim analysis covers 15 participants treated with DA-1726 and four on placebo, with an MRI substudy of 12 DA-1726-treated and four placebo participants. Dosing was titrated from 16 mg for four weeks to 32 mg for four weeks to the 64 mg target dose, given once weekly by subcutaneous injection.
| Endpoint, Day 109 | DA-1726 baseline | Change | Placebo-adjusted | p-value |
|---|---|---|---|---|
| Liver fat (MRI-PDFF) | 5.2% | All 5 with steatosis fell below 5% | −68% (GMR 0.32) | <0.001 |
| Body weight | 98.6 kg | −12.19 kg (−11.9%) | −13.5% | <0.001 |
| HbA1c | 5.79% | −0.44 pts (to 5.33%) | −0.24 pts | 0.006 |
| Waist circumference | 107.5 cm | −9.6 cm (−9.0%) | −9.1% | <0.001 |
Liver fat was measured by MRI-PDFF, with mean baseline of 5.2% in the DA-1726 arm versus 8.0% for placebo. The absolute treatment difference was −6.4 percentage points (p=0.007), and 10 of 12 treated participants, or 83%, achieved at least a 30% relative reduction, against none of four on placebo (p=0.008). Among the five with baseline steatosis, mean MRI-PDFF fell from 8.3% to 3.0%, and the highest baseline case dropped from 12.8% to 4.6%. Placebo liver fat rose 40.4% from its own baseline.
On body composition, visceral adipose tissue was 22% lower versus placebo, abdominal subcutaneous adipose tissue 19% lower, and total adipose tissue 19% lower. Thigh muscle volume was 9% lower versus placebo and total lean tissue 8% lower, against reductions of 19% to 22% across the measured fat compartments. For comparison, the S&P 500's year-to-date move is not disclosed in the report and no sector benchmark is supplied.
Analysis — what it means for markets / sectors / tickers
MetaVia is a clinical-stage company, so the equity case rests on pipeline probability rather than revenue. A 68% placebo-adjusted liver fat reduction with a p-value below 0.001 strengthens the probability weighting investors can assign to DA-1726, and the concurrent 13.5% weight loss and 0.24-point HbA1c improvement matter because they show the liver effect is not coming at the expense of metabolic benefit. The relatively preserved lean tissue, with muscle loss roughly a third to half the magnitude of fat loss, addresses a common objection to incretin-class drugs.
The limitation is sample size and duration. Twelve MRI-substudy participants and four placebo subjects over 16 weeks cannot establish durability, histologic benefit, or outcomes in patients with established disease. The company itself is planning a 48-week, non-biopsy Phase 2 study in up to 150 subjects with MetALD and ALD, targeted to begin in the second half of 2027, which signals that the confirmatory work is still ahead.
Exposure runs through the broader liver-disease and obesity drug complex, where competitors are also developing GLP-1-based and liver-directed assets. MetaVia's second program, vanoglipel (DA-1241), is being advanced in combination with resmetirom for MASH, with a Phase 2a proof-of-concept study targeted for the first half of 2027. Investors holding MTVA are effectively long two shots at the same liver-metabolic thesis; those short the space are betting on larger competitors' data reads and commercial scale.
Outlook — what to watch next
The first catalyst is full data from Phase 1 Parts 3A and 3B, which the company expects in the first quarter of 2027. That release should show whether the Day 109 liver and body-composition effects hold across the complete dataset rather than the interim cut.
The second is the start of the Phase 2 study in MetALD and ALD, targeted for the second half of 2027, in up to 150 patients and without a biopsy requirement. Whether MetaVia funds that trial alone or brings in a partner is not disclosed.
The third is the vanoglipel plus resmetirom Phase 2a proof-of-concept study in MASH, targeted for the first half of 2027. No price levels, moving averages or analyst targets appear in the report, so none are cited here.
Frequently Asked Questions
What does the 68% liver fat reduction actually measure?
It is the placebo-adjusted change in liver fat measured by MRI-PDFF, a magnetic resonance imaging method that estimates the percentage of fat in the liver. MetaVia reported a geometric mean ratio of 0.32 versus placebo at Day 109, with a p-value below 0.001. The absolute difference was −6.4 percentage points, and all five treated participants who started with steatosis fell below the 5% threshold.
What happens next for MetaVia and DA-1726?
MetaVia expects full Phase 1 Part 3A and Part 3B data in the first quarter of 2027. It is planning a 48-week, non-biopsy Phase 2 study of DA-1726 in up to 150 patients with MetALD and ALD, targeted to begin in the second half of 2027. The company did not disclose trial costs, financing plans or partnership terms for that study.
How safe was DA-1726 in this interim analysis?
No participant discontinued because of a gastrointestinal adverse event, and no new gastrointestinal events were reported after Week 9 through the Day 109 assessment. Nine of 15 DA-1726-treated participants reported gastrointestinal events, mostly mild to moderate, with vomiting in seven, nausea, diarrhea and dyspepsia in two each, and constipation and abdominal pain in one each. One serious event of severe vomiting was considered possibly related; the participant recovered.
Bottom Line
MetaVia's interim data show DA-1726 cut liver fat 68% versus placebo, but the 12-patient MRI substudy leaves Phase 2 to prove it.
